Cell sheets composed of monolayer cells have various applications in regenerative medicine. A cell sheet made from cardiomyocytes can be laminated to make thick myocardial tissue, but in order to engraft in vivo, it is indispensable to construct a vascular network to supply oxygen and nutrients to cardiomyocytes. is there. Therefore, in this study, we investigated the promotion of vascular network construction on cardiomyocyte sheets by local sustained release of vascular endothelial growth factor (VEGF) in vivo. A fiber mat that releases VEGF slowly was transplanted under the skin of a rat together with a six-layered cardiomyocyte sheet, and the tissue two weeks after the transplantation was collected to evaluate the thickness of the cardiomyocyte sheet layer. As a result, it was found that a remarkably thick tissue was engrafted and a blood vessel having a large cross-sectional area was constructed. This indicates that it is possible to construct a thick myocardial tissue having a vascular network in vivo by slowly releasing VEGF in the vicinity of the cardiomyocyte sheet.

Recently, there is a demand for external preparations for skin care with high moisturizing performance. The stratum corneum lipids of the human epidermis have a "lamellar" structure and retain a high barrier function. Lamellar means "layered", and a layered structure of water and oil exists between the stratum corneum cells. It is known that intercellular lipids not only exist as components, but also have a three-dimensional structure called lamella, which exerts a high barrier function. The lamellar structure is formed by a special amphipathic lipid molecule such as ceramide taking a liquid crystal structure together with an aqueous phase and an oil phase. This aqueous phase contains aqueous components such as amino acids called NMF (Natural Moisturizing Factor), which themselves have high moisturizing power and thus reinforce the barrier function of the skin. Recent advances in cosmetic emulsification technology have made it possible to artificially reproduce the three-dimensional structure of this lamella. The existence of the lamellar structure can be confirmed as a characteristic interference image under a polarizing microscope. In aged skin and rough skin, the barrier function of the skin may be deteriorated due to a lack of components constituting the lamella or a disorder of the lamellar structure itself. By applying an external preparation containing a three-dimensional lamellar structure composed of a water-soluble moisturizing factor, an amphipathic substance, and a lipid to the skin, it is possible to contribute to strengthening the barrier function of the skin. The purpose of this seminar is as follows. A highly moisturizing cream or gel is prepared using the ITO lamellar structure creation kit (ITO lamellar paste). Antioxidants such as fullerenes, vitamin C derivatives, astaxanthin, and coenzyme Q10 are encapsulated in the lamellar structure of the cream to prepare a highly functional preparation. Introducing clinical examples and basic data using nanosphere technology in which lamella paste is made into nano-sized fine particles.

In recent years, industrial technologies that utilize the properties of nanomaterials to bring many benefits to various fields of society have made remarkable progress, and the products of these technologies are already overflowing around us, but on the other hand, nanomaterials Is assumed to be a complex form of exposure to the human body, and due to its characteristics, concerns about health problems are also whispered. The future of nanotechnology depends on risk vs. the public interest, and unlike asbestos in the past, unlimited use without risk assessment is no longer allowed. Acne vulgaris is a multifactorial disease that develops at puberty. Coproporphyrin produced by Propionibacterium acnes (P. acnes) induces reactive oxygen species and promotes inflammation. In recent years, the several effects of ascorbic-acid (AsA) derivatives having an antioxidant activity for the treatment of acne vulgaris have been reported. Based on these reports, two AsA derivatives are recommended as option (C1) as cosmetic active agents for Japanese guideline for the treatment of acne vulgaris published by the Japanese Society of Dermatology in 2016. Glyceryl-octyl-ascorbic acid (GOVC) was developed for obtaining a significant effect on acne vulgaris. GOVC is a novel, amphiphilic stable AsA derivative with an antimicrobial activity. In this study, the stability, antioxidant effect, and antimicrobial activity by GOVC were investigated. In addition, clinical trials using topical GOVC were performed and its efficacy and adverse effects were evaluated on acne lesions.
(1) One percent GOVC aqueous solution showed a high stability of 98% or more without odor, discoloring, and precipitation after the storage test at 40 °C for 6 months. The residual ratio after autoclave treatment was more than 91%. (2) Radical scavenging activity of GOVC (0.1% solution) significantly decreased the amount of O2* down to 60.5% (n = 7, p <0.01) compared with that of the control. (3) In the chlorine radical scavenging activity test, GOVC (0.1% solution) significantly decreased the amount of chlorine radical down to 13.5% (n = 7, p ><0.01) compared to that of the control. (4) 0.1% GOVC showed an antimicrobial effect against P. acnes. (5) Topical GOVC was clinically effective for acne lesions. Using analyzing the number of automatically identified acne lesions, red inflammatory papules decreased significantly down to 55% (p ><0.05), and its area of postinflammatory erythema significantly decreased down to 32% (p >< 0.05).
A high stability, and an antioxidant and antimicrobial effects against P.acnes of GO-VC were confirmed. The clinical trials demonstrated that the number of inflammatory lesions and area of postinflammatory erythema were decreased significantly by topical GOVC. No adverse effects using topical GOVC were observed. Therefore, GOVC can be an alternative adjuvant available cosmetic agent for acne vulgaris.

However, it is difficult to achieve both stability and absorbability with conventional lipid-added derivatives, and GOVC has solved this difficult problem. GOVC is an amphipathic vitamin C derivative having a structure in which glycerin is bound to the hydroxyl group at the 2-position of vitamin C and octanol is bound to the hydroxyl group at the 3-position. The feeling of stickiness and tension, which was difficult with conventional vitamin C derivatives, has also been improved, and the common sense of vitamin C has been changed. Furthermore, GOVC was found to have strong antibacterial activity, and was found to have an inhibitory effect on not only P. acnes, which is the cause of acne, but also Escherichia coli and Staphylococcus aureus. Glyceryl octyl ascorbic acid (GO-VC) is a stable amphipathic AsA derivative in which glycerin and octanol are bound to the water hydroxyl groups at the 2- and 3-positions of vitamin C (AsA), respectively.
Our previous studies have already reported that GO-VC has a high stability, a fibroblast growth-promoting effect, and a type I collagen growth-promoting effect to improve wrinkles.
Since GO-VC binds octanol, which has antibacterial activity, it may exert antibacterial activity against P. acnes (P. acnes), which is involved in the development of acne vulgaris (acne).
This time, we investigated the effect of GO-VC on the growth of P. acnes and the effect of suppressing the production of melanin. We also conducted a clinical trial of GO-VC to examine its effect on acne, erythema, and color pigmentation. Furthermore, although conventional AsA and AsA derivatives have been reported to be used as a side effect to dry the skin abnormally, the moisturizing effect of GO-VC was also investigated. In the past, we have reported that the increase in AA radicals and reactive oxygen species (ROS) due to high-concentration vitamin C (AA) in UV-irradiated skin tissue may be due to iron proteins. Method: The subjects were 15 women aged 22 to 76 years. Abstract: Vitamin C (VC) is one of the most attractive vitamins in the fields of cosmetics and cosmetological skin. VC is a kind of water-soluble vitamin and is generally known as an antioxidant vitamin because it has a strong reducing power. It has been confirmed that the water distribution in the skin plays an important role in maintaining the homeostasis of the skin, and that changes in the state of the water distribution cause the production of inflammatory cytokines in the epidermis.
In addition, the homeostasis of the skin is impaired by the stimulation of the external environment, especially sunlight. Repeating such a condition leads to the formation of pigmented spots and wrinkles. It has been confirmed that VC has an action of promoting the synthesis of sphingolipid involved in the epidermal barrier function and an action of suppressing the production of inflammatory cytokines and chemokines derived from its antioxidant power. This paper describes the effects of VCs and VC derivatives on maintaining skin homeostasis.

Abstract: At last year's General Assembly, we reported that electroporation (electroporation) is cosmetically useful as an intradermal administration method of bFGF (Basic FGF), a human fibroblast growth factor that replaces injection therapy. "In vivo Gelatin: Collagen Biosynthesis Research Using Vitamin C Stabilizing Derivatives"

Abstract: Vitamin C is required for the formation of the three-helical structure of collagen. When vitamin C is deficient, the three-helical structure is not stably formed and is not secreted from the cell but decomposed inside the cell. In this way, the quality control mechanism of the molecular structure is assumed for collagen. However, it was shown that in type IV collagen, which is basement membrane collagen, a polypeptide chain having no three-helical structure is secreted extracellularly. Gelatin whose collagen structure is destroyed by heat treatment or the like is called gelatin, and it has been considered that gelatin does not exist in the living body because it is easily decomposed by protease. However, it was shown that a single-strand polypeptide chain having the same amino acid sequence as type IV collagen but not having a three-helical structure can exist in the living body. In this seminar, we will summarize the action of vitamin C in collagen biosynthesis and introduce our findings on the biosynthesis of gelatin in vivo and its physiological role.

Abstract: Various administration methods have been investigated for skin rejuvenation using the skin ulcer therapeutic agent Fiblast R (hereinafter referred to as bFGF). Although there are reports that administration is performed by subcutaneous injection, bFGF preparations are external preparations, and injection administration is not permitted at this time. We have obtained good results by administration by electroporation and ultrasonic iontophoresis without injection in cases for scar morphology improvement and skin rejuvenation, but we also administered by microneedle sheet. The usefulness was examined. Microneedle is a sheet in which extremely small needles made mainly of hyaluronic acid are lined up. When attached to the skin, the microneedles dissolve in the intradermal water, and the content components elute into the skin to break through the stratum corneum and deliver the active ingredients into the epidermis or dermis. The length of the needle can also be adjusted, and delivery from the stratum corneum to the inside of the dermis can be performed depending on the length. This time, using microneedle containing bFGF preparation and microneedle containing only hyaluronic acid, the effect of improving facial wrinkles was examined using a skin image analyzer Visia, and electroporation, ultrasonic iontophoresis and their usefulness were compared. Abstract: Vitamin C is essential as a cofactor for the hydroxylase of proline residue and lysine residue required for the maturation process in the synthesis process of collagen, which is a major component of skin, bone, tendon and the like. When we add ascorbic acid phosphate (Asc2-P, active-sustaining vitamin C) to the culture system of skin fibroblasts, liver cells, and osteoblasts, it activates cell proliferation, extracellular matrix metabolism, and is released. We found that three-dimensional tissues and organs can be regenerated from cells. Asc2-P promotes ECM maturation by activating the synthesis of collagen that forms the backbone of extracellular matrix (ECM). The formed ECM forms a microenvironment around the cell and gives the cell three-dimensional three-dimensional structure information to promote organ formation. In this talk, we will discuss various actions that cannot be explained by the conventional cofactor action of vitamin C, such as the recent anticancer action and antiviral action of vitamin C, as well as the action of Asc2-P in organogenesis.

Currently, stem cells are classified into several types such as embryonic stem cells (ES cells), somatic stem cells, and induced pluripotent stem cells (iPS cells), and application research for each stem cell to regenerative medicine is being conducted. Abstract: APPS (ascorbyl palmitate) is an interesting vitamin C derivative that has significantly improved skin permeability by binding lipids to ascorbic acid phosphate and modifying fatty acids to water-soluble vitamin C. In addition, TPS (Na tocopherol phosphate) is a vitamin E derivative whose permeability is improved by modifying fat-soluble vitamin E with water-soluble phosphoric acid. These antioxidant molecules have a hydrophilic and hydrophobic structure in the molecule. Therefore, it easily passes through the skin barrier layer and also retains an appropriate barrier property against intramolecular lipids. Therefore, it was considered that the property of solving the problem of dryness of conventional APS and the problem of stickiness of VCIP at the same time and having an extremely good feel after application is extremely useful as vitamin C for home care. Although these molecules contain lipids, water-soluble salt solutions can be easily prepared and iontophoresis is possible. We report that this external preparation containing APPS and TPS was applied to multiple cases that seemed to be effective in outpatient dermatology, and some findings were obtained.

Abstract: Acne vulgaris is a disease that occurs when various factors are intricately intertwined, so it is often experienced to be resistant to common treatments such as administration of antibiotics and vitamins, and once improved. Even if it looks like it, it recurs easily, and it is often difficult to cure it. We have performed complex treatments for acne vulgaris with antibiotics, retinoic acid, chemical peels, and vitamin C derivatives with very good results. These treatments can treat existing lesions and prevent the development of new lesions by effectively blocking the process of acne vulgaris development and exacerbation in multiple stages. The actual treatment of such complex acne vulgaris and the efficacy of the recently introduced novel vitamin C derivative APPS will be described in detail.
